Vertex Pharmaceuticals’ product portfolio is centered on medicines for cystic fibrosis, severe sickle cell disease and transfusion-dependent beta thalassemia, and acute pain. Its marketed cystic fibrosis medicines are ALYFTREK (vanzacaftor/tezacaftor/deutivacaftor), TRIKAFTA/KAFTRIO (elexacaftor/tezacaftor/ivacaftor and ivacaftor), SYMDEKO/SYMKEVI (tezacaftor/ivacaftor and ivacaftor), ORKAMBI (lumacaftor/ivacaftor), and KALYDECO (ivacaftor). In these regimens, CFTR potentiators such as ivacaftor and deutivacaftor increase the probability that CFTR channels open on the cell surface, while CFTR correctors such as lumacaftor, tezacaftor, elexacaftor and vanzacaftor improve protein processing and folding so more functional CFTR reaches the cell surface. ALYFTREK is described as a once-daily next-in-class triple combination for people with cystic fibrosis aged 6 and older with at least one F508del mutation or another responsive mutation; Vertex states it demonstrated non-inferiority to TRIKAFTA in ppFEV1 and improved sweat chloride versus TRIKAFTA, and is approved for 31 additional mutations not responsive to other CFTR modulators. TRIKAFTA’s U.S. label was expanded to include 94 additional non-F508del CFTR mutations, bringing the total to 272 approved CFTR mutations in the U.S.[8]

Outside cystic fibrosis, CASGEVY (exagamglogene autotemcel) is an ex vivo, non-viral CRISPR/Cas9 gene-edited cell therapy for patients aged 12 and older with severe sickle cell disease or transfusion-dependent beta thalassemia. The treatment process involves mobilizing and collecting a patient’s hematopoietic stem and progenitor cells, editing those cells at a manufacturing facility through a targeted edit in a non-coding intron of the BCL11A gene, then reinfusing the edited cells after myeloablative conditioning; Vertex describes the therapy as designed to raise fetal hemoglobin erythrocytes and potentially provide a one-time functional cure. JOURNAVX is a first-in-class oral pain signal inhibitor for moderate-to-severe acute pain in adults that is highly selective for NaV1.8 voltage-gated sodium channels. Financially, Vertex’s portfolio remains highly concentrated in TRIKAFTA/KAFTRIO, which generated $10.24 billion of $11.02 billion in 2024 total revenues, or about 93%, while other product revenues were $781.5 million, including $10.0 million from CASGEVY; Vertex reported no ALYFTREK sales in 2024.[12] [8]

Geographic presence

Vertex Pharmaceuticals’ reported sales base is concentrated in the United States and Europe. The company attributes net product revenues to countries based on customer location and reports geography at three levels: United States, Europe, and other markets outside the United States. In 2024, net product revenues were $11.0 billion, including $6.7 billion from the United States, $3.5 billion from Europe, and $0.9 billion from other international markets; this implies that about 61% of revenue came from the U.S. and about 39% from outside the U.S., with Europe representing the largest ex-U.S. region.[18][5]

Its principal executive offices are at 50 Northern Avenue in Boston, Massachusetts, and the company leases about 1.1 million square feet of office and laboratory space there across two buildings. Vertex also has established small-molecule manufacturing capabilities in Boston and owns about 213,000 square feet at a continuous manufacturing facility in Massachusetts. Beyond Massachusetts, the company identifies a research site in San Diego, California, and states that its European headquarters and European research facility are located in the U.K.; in addition, it leases roughly 850,000 square feet globally for logistics, laboratory, commercial, manufacturing, R&D support, and office uses.[19]

As of December 31, 2024, Vertex had approximately 6,100 employees, with about 5,100 based in the U.S. and about 1,000 based outside the U.S., indicating that its workforce remains predominantly U.S.-based even though it has material international commercial operations.[7] [18]

Business segments

Vertex Pharmaceuticals reports a single operating and reportable segment: pharmaceuticals, which therefore accounts for essentially 100% of the company’s business. Within that segment, the company’s focus is the discovery, development and commercialization of medicines for serious diseases; its commercial and pipeline activities are centered on cystic fibrosis, severe sickle cell disease and transfusion-dependent beta thalassemia, acute and peripheral neuropathic pain, APOL1-mediated kidney disease, IgA nephropathy and other autoimmune renal diseases and cytopenias, type 1 diabetes, myotonic dystrophy type 1, and autosomal dominant polycystic kidney disease.[25]

Vertex does not disclose multiple reportable business segments; instead, it disaggregates the pharmaceuticals segment mainly by product and geography. In 2024, TRIKAFTA/KAFTRIO contributed about 92.9% of net product revenue ($10.24 billion of $11.02 billion), while all other product revenues contributed about 7.1%, indicating that the single pharmaceuticals segment is still overwhelmingly driven by the cystic fibrosis franchise. By geography, 2024 net product revenue was approximately 60.7% from the United States, 31.3% from Europe, and 8.0% from other markets.[27] [25]

Company history

Vertex Pharmaceuticals was founded in 1989 by Joshua Boger and was incorporated in Massachusetts that year.[30][19] Its early commercial history included royalty income beginning in 1999 from GlaxoSmithKline’s sales of Agenerase, an HIV drug discovered under the companies’ collaboration, and the July 2001 acquisition of Aurora Biosciences.[31]

A major milestone came in 2011 with the U.S. approval of Incivek (telaprevir) for hepatitis C, followed in 2012 by FDA approval of Kalydeco, the company’s first cystic fibrosis therapy targeting the disease’s underlying cause.[34][35] Vertex expanded its cystic fibrosis franchise with Orkambi in 2015 and later with Trikafta in 2019, which became another landmark product for the company.[36][37]

In the late 2010s, Vertex also broadened beyond cystic fibrosis through new technology platforms, including a gene-editing collaboration with CRISPR Therapeutics and the 2019 acquisition of Semma Therapeutics to build a type 1 diabetes cell-therapy program.[38][39] In 2020, Reshma Kewalramani became Chief Executive Officer and President, marking an important leadership transition after Jeffrey Leiden’s tenure as CEO from 2012 through March 2020.[40][41] [30]
